What is the purpose of blank/background wells in the assay?

Blank or background wells are used to filter out artifacts, such as temperature and buffering capacity, that can affect oxygen and pH that are not due to changes in metabolism. Background wells can also be used to correct for any temperature fluctuations across the plate, used to diagnose potential thermal issues, and serves as a starting point for troubleshooting an assay with unexpected results.
