Activating Lasting Change in Childhood Cancer Treatments

In 2007, the Children’s Cancer Hospital Egypt (CCHE) 57357 was established to create a better tomorrow for children facing a devastating cancer diagnosis. At CCHE, Dr. Sameh Ali is Head of the Tumor Biology Research Program and concentrates his team’s efforts on mitochondrial physiology and pathophysiology. The team also focuses on as well as multifaceted characterizations of cellular function and death mechanisms. With a broad and detailed research scope, Dr. Ali needed powerful, highly sensitive instrumentation that was also versatile. He evaluated several options and ultimately selected the Agilent BioTek Cytation 5 cell imaging multimode reader. “We looked at all the application notes and publications using Cytation 5 and determined that it was best suited to fulfill our research and productivity needs,” he notes.

The Agilent BioTek local support staff helped Dr. Ali to configure a Cytation 5 specific to the needs of his laboratory. He credits the local team for providing timely and thorough responses to help guide his decision-making. In fact, he remains in touch with the Agilent BioTek experts for technical support and as more team members receive training.

The compact Cytation 5 is used for most of the laboratory imaging applications. The system is employed for measuring viability and cell death response following compound treatment, monitoring mitochondria and mitochondrial membrane potential, and determining calcium dynamics and reactive oxygen species (ROS). Accessories like atmospheric control and reagent injectors ensure that live cell and tissue slice experiments are time- and cost-efficient. 

In Dr. Ali’s lab, Cytation 5 multimode detection is used for optical enzyme activity assays, ELISAs, ROS release, calcium sequestration and release, and much more. Imaging and detection are both controlled and automated via Gen5 Software, which is also used for data and image analysis. Dr. Ali comments, “The combination of multi-well imaging and high-content analysis increases productivity and opens the horizon for experiments that would have been impractical to perform using conventional fluorescence microscopes.”
