Since 1966 Dako has produced polyclonal antibodies and our portfolio is constantly growing. Extensive knowledge of protein chemistry and immunochemistry, careful selection of animals for immunization, and optimal, long-term immunization schemes form the basis of our high-quality products.
Specificity
Monospecificity of Dako polyclonal antibodies is obtained by the use of highly purified antigens for immunization. Traces of unwanted antibodies are removed by liquid or solid-phase absorption.
Affinity-Isolated Antibodies
Dako’s antibodies are prepared by immuneaffinity chromatography, using antigens coupled to a solid matrix. The elution and adsorption techniques used guarantee antibodies of high affinity.
Immunoglobulin Fractions
The majority of Dako’s polyclonal antibodies are offered in the form of immunoglobulin fractions with a few exceptions mentioned under the individual product. The immunoglobulin fraction is prepared by salting out and ion exchange chromato-graphy. The elimination of bulk proteins gives a stable product with reduced background in gel precipitation techniques, and minimal non-specific reactions in other applications.
Low Batch-to-Batch Variation
Our batches of polyclonal antibodies consist of the pooled sera from a large number of animals. This method eliminates the presence of a single predominating atypical antibody and therefore leads to a minimal batch-to-batch variation.
Advantages of Rabbit Polyclonal Antibodies
Human antibodies reacting with rabbit immunoglobulins occurs rarely. Therefore, rabbit antibodies can be used without risk of non-specific binding even in very sensitive techniques. Polyclonals also help amplify signal from target protein with low expression levels by binding to more than one antibody molecule on the multiple epitopes. Additionally, polyclonals are more tolerant to minor changes in the antigen such as polymorphism, heterogeneity of glycosylation or slight denaturation, which is advantageous in certain types of assays.
Applications
Due to their high purity and avidity, Dako’s polyclonal antibodies are generally well suited for a variety of techniques such as nephelometry, turbidimetry, i munofixation, immunoblotting, and qualitative as well as quantitative gel precipitation techniques. Most of our antibodies have proven to be well suited for use in enzyme-linked immunosorbent assays (ELISA), chemiluminescence assays (CLIA) and lateral flow techniques.
