New Agilent Cary 8454 UV-Vis spectrophotometer delivers easy operation, ruggedness, speed, and compliance

The Agilent Cary 8454 UV-Visible spectrophotometer builds on our 30 years of experience in photodiode array (PDA) technology delivering an instrument that is easy to use, is ideal for routine applications, and meets the compliance needs of global pharmaceutical companies. This successor to the Agilent 8453 UV-Vis is the routine UV-Vis spectrophotometer of choice for QA/QC Pharma and time-based (kinetics) measurements in academia or chemical laboratories.
Designed for today’s laboratory
The Agilent Cary 8454 UV-Vis leverages the proven PDA detector used in Agilent’s Liquid Chromatography instruments, with pre-aligned light sources for trouble-free maintenance. Having very few moving parts, the Agilent Cary 8454 delivers the robustness and reliability needed in a QA/QC lab, maximizing uptime and lowering cost of ownership, and has the same small footprint as the 8453 UV-Vis, saving space in busy laboratories.
The instrument enclosure has been improved to incorporate hard-surface buttons for better chemical resistance and the convenience to start measurements away from the PC controller. The open sampling area improves ergonomics for accessory installation and makes sample presentation very easy, as well as eliminates errors that may occur from leaving the sample compartment door ajar on conventional UV-Vis instruments.
Faster performance
With the Agilent Cary 8454 UV-Vis, you collect a complete spectrum with every measurement, ensuring that you get the full picture instantaneously.
With each measurement, the PDA detector captures and automatically stores data across the full wavelength range from 190 to 1100 nm. Simultaneous multi-wavelength measurement is possible with the PDA technology of the Agilent Cary 8454 UV-Vis, and for applications such as quality control and quality assurance, the ability to quickly and completely assess a sample is a productivity benefit.
Proven compliance solutions
Agilent PDA spectrophotometers have been the pharmaceutical industry standard for decades, covering a range of applications from QA/QC, to component analysis and tablet dissolution. Agilent also offers a comprehensive portfolio of regulatory compliance services and support options to help achieve validation of the 8454 UV-Vis system and its components.
The Agilent UV-Visible ChemStation undergoes extensive validation during development, as documented by the Declaration of Validation shipped with each instrument. Two optional 21 CFR Part 11 compliant software tools are available – the workstation-based Security Pack software add-on, and the recently released Compliance Pack. The Compliance Pack software add-on enables a direct integration with the Agilent OpenLAB ECM solution for 21 CFR Part 11 compliance (Figure 3) providing:
- Customizable user roles and privileges
- A single audit trail that contains a record of all changes to methods and analysis of data for results files
- Electronic records and passwords given by the FDA 21 CFR Part 11

Seamless upgrade
The transition to an Agilent Cary 8454 UV-Vis is seamless and transparent. Agilent support tools simplify the transition from existing 845x systems, to eliminate productivity interruptions and ensure the continuation of existing standard operating procedures (SOPs). The Agilent Cary 8454 UV-Vis has been tested to ensure that standard methods give acceptable and equivalent results. By ensuring a seamless transition of methods from the 8453 UV-Vis to the new Agilent Cary 8454 UV-Vis spectrophotometer, the time-consuming and costly need for revalidation is minimized.

The new Agilent Cary 8454 UV-Vis spectrophotometer is an important addition to the Agilent molecular spectroscopy portfolio for routine UV-Vis and fluorescence applications (Figure 4).
